Introduction Sulfatase 2 (SULF2) is a member of sulfatase family and catalyzes the removal of 6-O-sulfate groups from the heparan sulfate (HS) disaccharide units of heparan sulfate proteoglycans (HSPGs). HSPGs are present on the cell surface as well as in the extracellular matrix and act either as sequestration sites for growth factor/cytokine ligands or as coreceptors in multiple signal transduction pathways. SULF2 mediated desulfation of HSPGs is proposed to decrease the affinity of HSPGs for a variety of signaling ligands, releasing them from sequestration sites and making them available for triggering signaling stimuli. SULF2 is localized in endoplasmic reticulum, Golgi apparatus and also on the cell surface, and is expressed at highest levels in tissues such as ovary, skeletal muscle, stomach, brain, uterus, heart, kidney and placenta. In its post-translational modification, the conversion to 3-oxoalanine (also known as C-formylglycine, FGly), of a serine or cysteine residue in prokaryotes and of a cysteine residue in eukaryotes, is critical for catalytic activity. SULF2 enzyme regulates the activities of a number of signal transduction pathways which are up-regulated in many tissues during development and disease.
