Integrins and RGD

Integrin proteins, a member of the cell surface receptor family, are overexpressed on the surface of cancer cells and are promising targets for macromolecular drugs and gene delivery for the treatment of cancer. Among them, αvβ3 integrins are well known for their role in cancer progression and are overexpressed in melanoma, glioblastoma, ovarian, breast and prostate cancer cells. Arginine-glycine-aspartic acid (RGD) peptides are the most common peptides responsible for cell adhesion to the extracellular matrix (ECM), are present in various species, interact with integrins with high affinity and are widely used as ligands for targeted drugs and gene delivery vectors.

Dendrimer-RGD Conjugation

In addition to applications in the chemical industry, dendrimers and their composites have made important advances in biomedical applications, including drug delivery, gene delivery, and MRI agents. Dendrimers are biocompatible, non-immunogenic, and easily introduce various chemical groups at the terminal and central positions. When combined with RGD peptides, dendrimers can target tumor cell surface integrins and mediate cell binding and adhesion to drugs for therapeutic or targeted delivery of imaging agents for more accurate cancer diagnosis.

Applications of Dendrimer-RGD Conjugation

RGD peptide-based dendrimers are widely used in biological research and medical devices, not only as multifunctional "smart" delivery materials, such as carriers for targeted drugs and imaging agents for targeted tumor cells, but also for tissue engineering to promote tissue regeneration.

Drug Delivery/Imaging Agent Delivery

  • Drug Delivery/Imaging Agent Delivery
    Dendrimers bound to RGD peptides act as nanocarriers to target drugs to specific cells, particularly cancer cells that overexpress integrin αvβ3. In addition, imaging agents targeting tumor cells can be delivered to enable more accurate diagnosis of cancer.

Gene Delivery

  • Gene Delivery
    RGD has been combined with a non-viral vector dendrimer to allow targeted delivery of genetic material to target cells, thereby improving transfection efficiency.

Tissue Engineering

  • Tissue Engineering
    As a ligand for ECM, RGD can control cellular responses to biological materials, such as attachment, proliferation, and differentiation. RGD-modified vascular tissue grafts can inhibit platelet adhesion, improve cell infiltration, and enhance endothelialization. Addition of RGD to cardiac scaffolds promotes cell adhesion, prevents apoptosis, and enhances tissue regeneration. RGD-coated bone tissue grafts can improve bone cell adhesion, proliferation, and survival. RGD can also be used for regeneration of retinal pigment epithelium.
