Gene therapy holds the promise of curing different types of human diseases and disorders. As an evolving field of medicine, gene therapy is still in its infancy and its main challenge is to find safe and effective vectors that can deliver genes to target cells. The vectors used for gene delivery can be divided into two categories: viral vectors and non-viral vectors. Safety issues are the main bottleneck of viral vectors for gene therapy, in addition, viral vectors tend to trigger a series of inflammatory or immune responses due to repeated administration, and in addition, viral vectors are difficult to mass produce. Therefore, viral vectors are not the first choice for gene delivery, and research should be focused on the development of non-viral vectors. Chitosan, one of the most reported cationic polymers of non-viral natural origin, is a linear polysaccharide consisting of randomly distributed 2-N-acetyl-2-deoxy-glucose (N-acetyl-glucosamine) and 2-amino-2-deoxy-glucose (glucosamine) residues with β-1,4-linkage. As a non-viral carrier for gene therapy, chitosan is widely used for drug and gene delivery because of its biodegradability, biocompatibility, non-allergenicity, mucoadhesive property and strong affinity with DNA.

Dendrimer-Chitosan Conjugation Improves Transfection Efficiency

Non-viral vectors such as cationic lipids and cationic polymers are widely used as gene delivery vectors due to their low immunogenicity, versatility of chemical structure and low production cost. When non-viral vectors are used for gene delivery, it is difficult to produce systemic therapeutic effects due to low transfection efficiency. Chitosan is one of the promising non-viral vectors for gene delivery due to its natural origin, biodegradability, biocompatibility, and very low toxicity. However, its poor water solubility and low transfection efficiency are the main drawbacks when used as non-viral vectors for gene therapy. Dendrimers are relatively new class of synthetic, highly branched, nanospherical well-defined architectures with precise molecular weight, multivalent functionalization sites and low dispersity index macromolecules. When the dendrimers are conjugated to chitosan via N-maleation, the DNA complexation ability is significantly increased, allowing the formation of spherical nanoparticles with pDNA. It also exhibits protection of DNA from nuclease degradation, high transfection efficiency, low cytotoxicity, and good hemocompatibility.
