Introduction into Chemical Glycosylation

Chemical glycosylation is the coupling of two monomeric sugar units by the formation of a new linkage (known as the glycosidic bond) involving a glycosyl donor to a glycosyl acceptor. When both the acceptor and donor are sugars, the product is an oligosaccharide. Chemical glycosylation reactions occur under appropriate reaction conditions by using an activator to initiate the coupling of the glycosyl donor and acceptor. The activator commonly used for glycosylation is a Lewis acid, which enables the leaving group at the anomeric position to leave and results in the formation of the oxocarbenium ion. Since a new stereogenic center is created at the isomeric position of the glycosyl donor, the reactions often result in a mixture of products.

In glycosylation reactions, the glycosyl donor is some sugar with a suitable leaving group in the isomeric position. It is activated under the appropriate reaction conditions and then forms a carboxyl group that is eliminated, leaving an electrophilic isomeric carbon. The glycosyl acceptor is some sugar with an unprotected nucleophilic hydroxyl group that attacks the carbon of the oxocarbenium ion formed during the reaction and allows the formation of a glycosidic bond.

Dendrimers Glycosylation

As an amazing chemical molecule, dendrimers are the fourth class of polymers that have made great strides since their inception. Primarily used in the biomedical field, dendrimers are valuable in diagnostics and therapeutics due to their ability to improve drug solubility, absorption, bioavailability, and targeted drug distribution. In contrast to conventional polymers, dendrimers are characterized by branched repeating units that appear in a focal point and possess many exposed anionic, neutral, or cationic terminal features on the surface, thus behaving as hydrophilic or hydrophobic compounds. They are nanoscale molecules that are radially symmetric, spherical, monodisperse and homogeneous. Due to their size, dendrimers are used in nanomedicine research.

The ends of dendritic polymers can be modified by small molecules (including ions, drugs, and biomolecules) that alter the physicochemical, reactive, kinetic, and biological properties of the dendrimer ground. Properties can also be altered by undergoing acetylation, PEGylation, fluorination and glycosylation. When glycosylation of dendrimers occurs, it removes the surface cationic charge, which not only reduces cytotoxicity but also has immunomodulatory and anti-vascular production properties.
