DMPK studies the Absorption/Administration, Distribution, Metabolism, and Elimination/Excretion (ADME) properties of chemical compounds to assist the selection of optimal drug candidates. Definition of ADME
ADME is an abbreviation for "absorption/administration, distribution, metabolism, and elimination/excretion", which describes the disposition of a pharmaceutical compound within an organism. In vitro and in vivo DMPK/ADME studies enable researchers to identify drugs with high potential.
Absorption defines the movement of a drug being taken up the target cells. It critically determines the compound's bioavailability. Chemical compounds differ from each other in physiochemical properties (e.g., molecular weight, solubility) and biochemical characteristics (e.g., membrane permeability, metabolism), which may affect the absorption of a drug.
Distribution refers to the transportation and distribution of the drug after been absorbed. Factors affecting drug distribution include molecular size, polarity, binding target, and drug-drug interactions.
Metabolism refers to the breakdown of the drug, which plays a major role in both efficacy and safety. Most metabolism of small-molecule drugs takes place in the liver by redox enzymes (i.e., cytochrome P450 (CYP450) enzymes).
Excretion refers to the removal of the chemical compounds and their metabolites, which usually through urination.
